The Effects of Topical Nicardipine on Cerebral Pial Vessels in Rabbit


Abstract
Topical applications with normal saline and nicardipine solution of various concentrations on cerebral pial vessels in rabbits were performed in a randomized in vivo study and the reactions of pail vessel were studied through the bilateral
craniectomized areas.
Physiological parameters (PaO2, PaCO2 blood pH and systolic blood pressure) were not significantly changed during all experiments.
Diameter variation of 90 arteries with a range of 60-210 §­diameter and 76 veins with a range of 70-230 §­diameter were continuously analyzed using micrometer eyepiece on operating microscope.
1. Topical application with normal saline on cerebral pial arteries did not significantly change pail arterial diameter compared with the arterial diameter in the resting state (p>0.05).
Nicardipine solution with 50 and 100 §­% concentration did not significantly change the pail arterial diameter compared with normal saline(p>0.05), but 200, 400 and 800§­% nicardipine solution induced the dilatation of large and small pial
arteries
significantly by dose-dependent manner and time course (p<0.05, p<0.01), but the smaller arteries were dilated more than the large garteries(p<0.05).
2. In venous segments, topical application of the normal saline induced no significant pial vein dilatation compared with the resting state (p>0.05).
Pail veins exhibited only minor reaction by topical application of nicardipine solutions and then pial veins did not significantly dilated by topical administration of nicardipine solution with a range of this experiments.
Thus it may be suggested that topical application of higher concentration nicardipine solution (concentration >200 §¶%) induce dilatation of pail arteries with dose-dependent manner and time course. And then nicardipine might be used for cerbral
sichemic insult.
